First of all, here's how my screen currently looks (not all of the uimods are shown, but the ones listed below are active):

I decided on CTMod after using Cosmos and finding it too bulky. Also, I didn't like their force bow hooks put in for developers to be l33t. Anyways, I decided to bulk up my ui after the fact anyways. Here's what I'm using

All In One Inventory

-self-explanatory... gives you a Diablo style bag so you don't have to spend time micromanaging your bags only to have an empty space in your potions bag when you're trying to pick up vendor trash.
CooldownCount:

-instead of a graphical representation, this pops numbers letting you know the amount of seconds (or minutes) until your spell is ready to be cast again.

Rest XP To Go:

-lets you know how much "Rested" xp you have stockpiled in a quick glance. Also updates realtime when at an inn or in town.

-manages a minidatabase of items you've brought to the vendor (not necessarily sold) so you can decide on trashing an item in the future, or holding onto it to sell later.

-probably my favorite mod, this gives you the entire text combat window presented in an easy to read graphical format. when you're 1v1'ing there's a seesaw diamond which plays tug of war on the top of Showdown from left to right depending on who's currently "winning" (ratio of hp's). Also it lists your last 6 hits (or misses) and damages, as well as showing the same for the enemy. Doens't work in PVP... yet, but PVE is enough reason to get this mod.
Telos' Clock

-get the fuctionality of Cosmos' clock, without Cosmos. Comes with a time till level estimator, % run speed speedometer, and of course clock.

Some cry cheater when they hear what this mod does... basically it uses a potion, or healthstone, or mana crystal automatically when you get to a specific programmable % level. I love it, and Blizzard says it's legal for now (got this from the official UI customization thread).

Berserkplosion

-upon 20% health, this nifty mod pops up a full screen invisible button as well as flashing a little "Beserkplosion!" over your head to let you know you can cast the troll racial beserk ability. Click ANYWHERE on the screen to activate it.

-pops up when in battle, and gives you stats on your different attacks, and has pulldown menu to look at stats of a particular ability/spell. Comes with ctmod, but just in case you want it without downloading all of ctmod, here it is.
Guild On Tooltip:

-when you mouseover a person, you get their guildname as well, so you don't have to run right up them to read it. Also works on alliance, so you can peek over a hill, mouseover a gnome, and then read what guild he's in before you decide to gank them.
Scrolling Combat Text:

-adds Final Fantasy scrolling numbers above your head (just like it is already for enemies) so you don't have to look at the top left of your screen to see how much you're getting hit for. Allows you to keep your eyes on the action, and not worry about anything else. This pretty much is an either or both coupled with Showdown, but I just love being able to see the same information presented in different ways.
All these mods play nice together, and they all help me play better as well. I know people like to play vanilla, but here's what you're missing out on.
